Cardiovascular Disease Metabolomics Service

Cardiac metabolic disorders underlie most cardiovascular diseases. Metabolomics is a powerful tool to reveal changes in overall and region-specific metabolism in cardiovascular disease states. Metabolomic analysis can reveal metabolic abnormalities in the etiology of cardiovascular disease (CVD) and thus help identify novel biomarkers.

The human heart beats an average of 2.5-4 billion times in a lifetime, yet despite breaking down 20 times its weight in ATP per day, it can store only enough energy to sustain a few heartbeats. Therefore, the heart must adapt to changing nutritional availability and physiological demands to meet ongoing energy needs. In fact, the heart can produce ATP from a variety of sources including carbohydrates, lipids, lactic acid, amino acids and ketone bodies. Most cardiovascular diseases involve cardiometabolic disorders due to the critical need for a continuous energy supply.

In addition, metabolic disturbances in diseases such as diabetes directly affect cardiac metabolism. Cardiac disease itself can also affect systemic metabolism. Thus, systemic and cardiac metabolic disorders may trigger a vicious cycle leading to cardiovascular disease.

Creative Proteomics provides metabolomic analysis solutions for cardiovascular disease, combining targeted and untargeted metabolomics technologies with multivariate analysis to explore disease mechanisms, screen biomarkers, and facilitate drug development and disease diagnosis and treatment.

Strategies for Metabolomic Analysis of Cardiovascular Diseases

There are two main approaches to identify and analyze metabolites using metabolomics: untargeted metabolomics and targeted metabolomics.

Untargeted metabolomics is used to assess metabolites in samples and can reveal new or unanticipated changes in metabolites. Its main advantage is that it provides an unbiased approach to analyze the relationship between interrelated metabolites from multiple pathways. Metabolites in samples can be evaluated in detail, revealing a wide range of metabolite classes.

Targeted metabolomics measures the concentrations of a predefined set of metabolites. A standard curve for the concentration range of the target metabolite is prepared, resulting in accurate quantification. This type of analysis can be used to obtain the concentration of an exact metabolite determined by untargeted metabolomics, thus providing analytical validation. Targeted metabolomics offers higher sensitivity and selectivity than non-targeted metabolomics.
