A steroid is an organic compound with four rings arranged in a specific configuration, such as dietary lipid cholesterol, estradiol, testosterone and dexamethasone. Steroid biosynthesis (steroidogenesis) begins with the transfer of free cholesterol from intracellular stores into the inner mitochondrial membrane, where it is transformed into pregnenolone. The conversion of cholesterol to pregnenolone is the rate-limiting step of steroidogenesis and occurs in the single substrate-binding pocket of the cholesterol side-chain cleavage enzyme.
